New invention: Fabrication of polystyrene micro carriers with halal gelatin coating.

Abstract

 

In this work polystyrene (PS) microspheres were fabricated by using oil in water (o/w) emulsification coupled with solvent evaporation technique. In order to render the microspheres suitable for cell attachment and gelatin immobilization on their surfaces PS microspheres were loaded with active oxygen species such as carbonyl (CO) Hydroxyl (OH)  and Carboxyl (COOH) functional groups by using an ultraviolet / ozone (UVO) system. To mimic the extracellular matrix, PS microspheres were further modified in which halal bovine gelatin were covalently immobilized on their surfaces by using zero length  cross-linker reagents , EDAC/NHS. The morphology and composition of the gelatin coated PS microspheres were characterized by using laser particle size analyzer,  scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX) , Fourier transform infrared spectrum (FTIR) , water contact angle (WCA), toluidine blue O (TBO) and gelatin essays while their cellular bioactivity were assessed by growing Vero cells in suspension culture.  Results revealed that cells could attach , proliferate and spread on these PS micro carriers and the peak cell density achieved was highly comparable to culture that uses commercial micro carriers, Cytodex 1 and 3.
